Molecular marker heterozygosities and genetic distances as correlates of production traits in F1 bovine crosses Genet. Mol. Biol.
Tambasco-Talhari,Daniella; Alencar,Maurício Mello de; Paz,Cláudia Cristina Paro de; Cruz,Geraldo Maria da; Rodrigues,Armando de Andrade; Packer,Irineu Umberto; Coutinho,Luiz Lehmann; Regitano,Luciana Correia de Almeida.
Several studies have investigated the relationship between heterozygosity, genetic distance and production traits. The objective of the present study was to evaluate the influence of the degree of heterozygosity and genetic distance on growth, carcass and reproductive related features in F1 bovine crosses. We tested 10 polymorphic markers in 330 purebred cattle (Nelore, Canchim, Aberdeen Angus and Simental) and 256 crossbred cattle belonging to four crossbred groups. Individual heterozygosities (Hi) and multilocus genetic similarity (Dm) were estimated and used in correlation analysis against individual phenotypic measurements. Significant (p < 0.05) Hi effects occurred for birth weight, 15 to 18 month weight, hot carcass weight and longissimus rib eye...

Molecular markers to study the variability within the Eragrostis curvula complex Phyton
Zappacosta,D; Meier,M; Carrera,A; Pacheco,G; Cardone,S; Selva,JP; Echenique,V.
Weeping lovegrass is well adapted for forage production and useful for soil conservation in semiarid regions, constituting a morphologically diverse group. Diploid genotypes are unfrequent and reproduce sexually, whereas the tetraploids and plants of higher ploidy levels reproduce by apomixis. In the present work RAPD, AFLP and EST-SSR were used in order to assess the reproductive mode through progeny tests, to determine intracultivar homogeneity or seed purity, to establish genetic relationships among the cultivars within the complex and to characterize the new materials obtained by our group. Eight commercial cultivars and three new plant materials were analyzed. Uniform and variable patterns were observed in progenies of apomictic and sexual plants,...

Molecular Tools for Rapid and Accurate Detection of Black Truffle (Tuber melanosporum Vitt.) in Inoculated Nursery Plants and Commercial Plantations in Chile Chilean J. Agric. Res.
Cordero,Cecilia; Cáceres,Pablo; González,Gloria; Quiroz,Karla; Carmen,Bravo; Ramírez,Ricardo; Caligari,Peter D.S; Carrasco,Basilio; García-Gonzales,Rolando.
Truffle (Tuber melanosporum Vitt.) culture is an agroforestry sector in Chile of increasing interest due to the high prices that truffles fetch in the national market and the recent evidence that its commercial production is possible in Chilean climatic and soil conditions. In this study, the efficiency of three methods of DNA extraction from a mix of 5 g of soil and roots from both nursery and field plants of Quercus ilex L. mycorrhized with T. melanosporum were evaluated, and a simple and reproducible protocol was established. Detection of T. melanosporum was performed by the technique of cleaved amplified polymorphic sequence (CAPS) from amplicons generated with the primers ADL1 (5´-GTAACGATAAAGGCCATCTATAGG-3´) and ADL3 (5´-CGTTTTTCCTGAACTCTTCATCAC-3`),...

New method for early detection of two random amplified polymorphic DNA (RAPD) groups of Staphylococcus aureus causing bovine mastitis infection in Paraná State, Brazil BABT
Gonçalves,Dicezar; Gabriel,Jane Eire; Madeira,Humberto Maciel França; Schühli,Guilherme Schnell e; Vicente,Vânia Aparecida.
The aim of this work was to develop a fast and accurate molecular approach to allow early detection of two RAPD groups of S. aureus causing bovine mastitis. Seventy five S. aureus isolates from infected animals were characterized by RAPD. Genomic fragments isolated from the unique bands present in either group were cloned and sequenced. Based on the DNA sequences, specific primers were designed to allow for the simultaneous detection of either group by multiplex PCR of S. aureus DNA isolated from clinical and subclinical bovine mastitis. Results showed that these proposed primers set could be used to detect various clinical and subclinical S. aureus isolates as well as the detection of the microorganism in bulk milk. Their use as a specific method for...

Nucleotide diversity of a major carotenoid biosynthetic pathway gene in wild and cultivated Solanum (Section Lycopersicon) species Braz. J. Plant Physiol.
Araújo,Ana H.; Fonseca,Maria Esther de N.; Boiteux,Leonardo S..
A polymerase chain reaction (PCR)-based approach was employed to isolate putative alleles of the chromoplast-specific lycopene beta-cyclase (CYCB) gene from wild and cultivated tomatoes [Solanum (Section Lycopersicon)]. The objective of this work was to establish an effective PCR protocol by testing DNA samples from distinct germplasm accessions with a primer pair designed to selectively target conserved regions present in the available CYCB sequences. This PCR optimization allowed the amplification of 1219 out 1666 bp of the gene in six taxa: S. cheesmaniae, S. peruvianum, S. neorickii, S. pennellii, S. pimpinellifolium and S. lycopersicum. Sixty-three mutation sites (31 transitions, 18 transversions and 14 single base deletions/insertions) were detected...

Carmo,Fausto Moreira da Silva; Polo,Érico Macedo; Silva,Marcos Antônio da; Yazbeck,Gabriel de Menezes.
Abstract: The objective of this work was to evaluate 41 microsatellite markers for heterologous amplifications in piracanjuba (Brycon orbignyanus). Some markers were tested for the first time. Loci were optimized for PCR conditions and applied to a sample of 49 individuals. Thirty-one loci resulted in PCR product formation, whereas ten loci yielded intelligible polymorphic patterns in the evaluated sample and can be used for amplifications in this species. From the evaluated markers, four loci (BoM1, BoM13, Bh6, and Bh16) are valid to be applied in the study of piracanjuba.

Matos,Rafael de; Resende,Juliano Tadeu Vilela de; Zeist,André Ricardo; Corte,Lígia Erpen-Dalla; Da-Silva,Paulo Roberto; Zeffa,Douglas Mariani.
ABSTRACT Knowledge of the combining ability of genotypes, genetic diversity, and heterosis play the paramount role in obtaining double-cross hybrids with high productivity. This study was aimed at evaluating these parameters in tomato genotypes using partial diallel crosses among the commercial single-cross hybrids, which verified the possibility of using double-cross hybrids for commercial purposes. Tomato genotypes included 15 double-cross hybrids, their genitors, and two commercial genotypes as checks. Of the fifteen double-cross hybrids of tomato, two crosses exhibited superiority, such as Aguamiel × Compack and Dominador × Compack, mainly for the total and commercial fruit yields. The phenotypic expression of the trait is controlled by the genes with...

Pilocarpine content and molecular diversity in jaborandi Scientia Agricola
Sandhu,Sardul Singh; Abreu,Ilka Nacif; Colombo,Carlos Augusto; Mazzafera,Paulo.
Pilocarpine is an imidazol alkaloid exclusively found in Pilocarpus genus and P. microphyllus accumulates its highest content in the leaves. There is no report in the literature on the variability of the pilocarpine content in this genus. A population of 20 genotypes of P. microphyllus from the state of Maranhão, Brazil, was analyzed for Random Amplification of Polymorphic DNA (RAPD) markers and pilocarpine content. Although it was not possible to establish any correlation between these features, the absence or presence of some markers could indicate in some genotypes a possible association with the content of the alkaloid.

Polymorphism of the grasshopper Schistocerca pallens (Thunberg) (Orthoptera: Acrididae) and its natural pathogen Metarhizium flavoviride Gams & Rozsypal (Hyphomycetes), revealed by RAPD analysis Anais da SEB
Silveira,Eni B.; Al-Janabi,Salah M.; Magalhães,Bonifácio P.; Carvalho,Luiz J.C.B.; Tigano,Myrian S..
The genetic variability of 27 individuals from two distinct populations (Northeast and Central Brazil) of the grasshopper Schistocerca pallens (Thunberg) and eight isolates of its natural pathogen, the fungus Metarhizium flavoviride Gams & Rozsypal, was investigated using RAPD analysis. For the grasshoppers, ten different 10-mer oligonucleotide primers of arbitrary sequence were selected for analysis, resulting in 79 scorable binary characters. The program RAPDDIP applied to S. pallens revealed nucleotide diversity of 2.3 and 2.2% for the populations from Rio Grande do Norte (Northeast Brazil) and Federal District (Central Brazil), respectively. These values indicate the presence of high genetic variability within these populations. Conversely, the...

Predicting performance of soybean populations using genetic distances estimated with RAPD markers Genet. Mol. Biol.
Barroso,Paulo Augusto Vianna; Geraldi,Isaias Olívio; Vieira,Maria Lúcia Carneiro; Pulcinelli,Carlos Eduardo; Vencovsky,Roland; Dias,Carlos Tadeu dos Santos.
In order to verify whether genetic distance (GD) is associated with population mean (PM), genetic variance (GV) and the proportion of superior progenies generated by each cross in advanced generations of selfing (PS), the genetic distances between eight soybean lines (five adapted and three non-adapted) were estimated using 213 polymorphic RAPD markers. The genetic distances were partitioned according to Griffing's Model I Method 4 for diallel analysis, i.e., GDij = GD+ GGDi+ GGDj + SGDij. Phenotypic data were recorded for seed yield and plant height for 25 out of 28 populations of a diallel set derived from the eight soybean lines and evaluated from F2:8 to F2:11 generations. No significant correlation for seed yield was detected between GD and GV, while...
